Among the 50 entrants in the competition, Glycotope was selected as the winner for the innovative nature and commercial impact of its GlycoExpressTM technology of human cell lines to develop and manufacture biopharmaceuticals with an improved glycosylation leading to potentially significant therapeutic and economic benefits. Dr. Steffen Goletz, CEO, CSO & Founder of Glycotope is “proud, that in winning the award, Glycotope is joining technological leaders such as Thermo Fisher Scientific Inc., BASF, Novozymes A/S, DSM or Dow Pharmaceutical Sciences Inc.”

About Glycotope:

GLYCOTOPE, founded in 2001 in Berlin, focuses on the optimization and humanization of therapeutically relevant sugar structures on biopharmaceuticals known as glycosylation and on the development of innovative antibodies targeting glycostructures on cell surfaces. Biologics developed in Glycotope´s GlycoExpressTM technology of glycoengineered human cell lines receive a fully human and optimized glycosylation, leading to a largely improved activity, an elongated half-life time and/or reduced immunogenicity and hence important therapeutic benefits for a significantly increased number of patients. In addition, the GlycoExpress platform is superior in productions yields, product quality and reproducibility compared to other production platforms.

Glycotope´s product portfolio now encompasses a variety of antibodies and non-antibody biologics, 4 of which are currently in the clinical stage. This pipeline includes antibodies against novel targets for cancer therapy (e.g. GT-MAB 2.5-GEX™) and significantly improved versions of antibodies as well as other already marketed therapeutic proteins, so called Biobetters.

Together with Glycotope-Biotechnology in Heidelberg, Glycotope evolved into a leading integrated glycobiology company, covering all phases in drug development such as lead discovery and optimization, preclinical and process development, GMP production and clinical development. With more than 140 employees, the group has grown to one of the largest Biotech organisations in Germany.

About the CPhI Innovation Award

The internationally acclaimed CPhI Innovation Awards were established to recognise, celebrate and honour those companies and organisations that are breaking new ground in the pharmaceutical, contract services, packaging and biopharmaceutical.
Berlin, September 19th, 2011

Berlin, Germany, September 19, 2011 - Glycotope GmbH, a leading company in the field of glycomics, announced today the enrollment of the first patients in a phase I dose finding trial to evaluate safety and tolerability of TrasGEX™. TrasGEX™ is a biobetter of the marketed Trastuzumab / Herceptin™ targeting the HER2-receptor, commonly overexpressed in a variety of cancers. TrasGEX™ has been glycooptimized to yield a manifold higher anti-tumor activity and is supposed to be suitable for the treatment of an enlarged patient spectrum. It was developed using GlycoExpress™, the company’s development and production platform based on glycoengineered human cell lines for biopharmaceuticals with fully human and optimized glycosylation.

“We are delighted to see our 4th glycooptimized biopharmaceutical product in clinical trials”, says Dr. Steffen Goletz, CEO, CSO and Founder of Glycotope. “After CetuGEX™ and PankoMab-GEX™, TrasGEX is our 3rd anti-cancer antibody which entered clinical trials in time. GlycoExpress has been proven to be the broadest glycooptimization technology not only for antibodies but also for most other glycoproteins. Our glycooptimized Follicle-Stimulating Hormone FSH-GEX™ has already completed clinical phase Ia with very exciting results. Currently we are advancing a broad range of pre-clinical products in various therapeutic areas”.

Berlin, May 18th, 2011

Berlin, Germany, May 18th, 2011/ B3C newswire / – After the two monoclonal antibodies GT-MAB 2.5-GEX™ and CetuGEX™, the German biotech company Glycotope has recently received approvals to initiate clinical trials for its first glycooptimized non-antibody protein therapeutic FSH-GEX™. FSH-GEX™ is a follicle-stimulating hormone which will be developed for in vitro fertilisation and the treatment of anovulatory infertility. It is produced recombinantly with a fully human glycosylation. In April Glycotope has enrolled the first subjects into a phase I clinical trial in the Netherlands to evaluate the safety, tolerability, dosing requirements and pharmacokinetics of the product.

Based on GlycoExpress™, Glycotope´s development and production platform of glycoengineered human cell lines, FSH-GEX™ combines into one single product the safety advantages of a recombinant origin with the fully human nature of pharmaceuticals derived from urinary sources plus the glycooptimization of the molecule.

“After having 2 antibodies in clinical trials and a third antibody approved by the competent authority, starting the clinical phase for FSH-GEX™ is an important milestone for the company, as it is our first glycooptimized non-antibody product in the clinic, validating the broad applicability of GlycoExpress™ to not just antibodies but almost all glycosylated proteins”, states Dr. Steffen Goletz, CEO, CSO and Founder of Glycotope. “In addition, we are expecting that the first patients for TrasGEX™, our glycooptimized Trastuzumab, will be enrolled within the next weeks. We then will have successfully completed our ambitious plans to bring four glycooptimized biopharmaceuticals into the clinic within a very short time”.

Berlin, Heidelberg, April 5th, 2011

Berlin, Heidelberg, April 5th, 2011/ b3c newswire / -Glycotope Biotechnology GmbH in Heidelberg, Germany, a full subsidiary of Berlin based Glycotope GmbH, has initiated the setup of further clean room facilities as a first step in the expansion of its GMP-production capacities. By mid 2011, the company will thus be able to produce 4kg of recombinant antibodies per year and up to 11 kg upon completion of works towards the end of 2011. In sum, additional 1400 m² of facility and office space will be created.

The delighted Dr. Steffen Goletz, founder and CEO of Glycotope GmbH comments: “In particular the expansion of our DSP (Downstream Processing) capacities now places us among the top providers of GMP-production for recombinant proteins and in particular antibodies in Germany”.

About Glycotope and GlycotopeBiotechnology:

Glycotope-Biotechnology, a full subsidiary of Berlin based Glycotope GmbH was founded upon the acquisition of the GMP-production unit of former Orpegen Pharma GmbH, together with the transfer of manufacturing lines of former Avesta GmbH in Kleinmachnow (Berlin). With combined more than 140 employees, the Glycotope Group now has grown to one of the largest Biotech organisations in Germany. Glycotope´s product portfolio now encompasses a variety of antibodies and non-antibody biologics, 3 of which are currently undergoing clinical trials. The pipeline includes both new antibodies for tumor therapy (e. g. PankoMab) as well as significantly improved versions of already marketed protein therapeutics (BioBetters).

Glycotope´s underlying GlycoExpress™ technology of human cell lines facilitates the optimization of previously disregarded carbohydrate structures found on protein-based drugs (glycosylation). Thus biologics developed in GlycoExpress™ receive a fully human and optimized glycosylation, leading to a largely improved activity, an elongated half-life time and reduced immunogenicity and hence important therapeutic benefits for a significantly increased number of patients.

Berlin, 29 Oct. 2007
Jossa Arznei GmbH in Munich has acquired a participatory interest in GLYCOTOPE GmbH in Berlin in the context of a capital increase and has undertaken to provide EUR 40 million during the first stage. The money is to finance the clinical development of three drugs up to the final clinical phases and the admission of in-house production cell lines. The investor has also undertaken to provide additional funds to the biotech company in Berlin-Buch in a second stage for the extension and further development of the company’s technology and product pipeline along the value-added chain.

GLYCOTOPE was founded in 2001 by the biologist and biochemist Dr. Steffen Goletz and seed phase investor Eckert Consult Strategieberatung und Kapitalbeteiligungsgesellschaft mbH. The company operates on glycosylation structures on proteins, a relatively young sector in biotechnology. Its proprietary technology for optimization of glycosylation structures on proteins and biopharmaceuticals production permits making existing and new drugs more effective and tolerable in the human body, which is of considerable economic importance for new as well as improved versions of protein-based drugs (2nd generation biopharmaceuticals).

The GLYCOTOPE GmbH product range features a number of antibodies and other proteins. They include both proprietary antibodies for tumour therapy (PankoMab, KaroMab) and significantly improved versions (2nd generation) of antibodies and other therapeutic proteins already on the market. The innovative company’s base technology supports the development of new active substances and marked improvement of the glycosylation structures and optimization of their effectiveness.

„The fresh capital helps us improve our financial resources dramatically, while also bringing us a strong and experienced partner. This will enable us to accelerate the development of our projects considerably and expand our glycosylation technology platform,“ said the management team comprising Dr. Steffen Goletz, Dr. Franzpeter Bracht and Dr. Hans Baumeister.

Since its foundation in 1982 ORPEGEN had internationally developed many close and trustful business relations with clients from pharmaceutical industries, mid sized companies and younger start-ups, as well as research institutions, clinics and private individuals. These lasting good cooperations with our clientele have evolved not only from contract R & D, peptide chemicals supply and biotechnological contract development & manufacture, but also from our providing to international partners our innovative novel diagnostics, which for the first time allow the quantification of human cellular immune responses.

Glycotope is maintaining these outreaching business relations and is pleased and privileged to name a few of our business clients:

TrasGEX is an improved version of the marketed blockbuster drug Trastuzumab / Herceptin (Roche, Genentech, Chugaj). Trastuzumab is targeting the HER2-receptor, commonly overexpressed in a variety of cancers.

Herceptin has been approved for the treatment of HER2 positive breast cancer and HER2 positive stomach cancer.

Glycotope´s TrasGEX has been glycooptimized in Glycotope´s fully human development platform GlycoExpressTM. As a result, TrasGEX shows a significantly improved ADCC-activity, in particular among all allotypes of the FcγIIIa-receptor mediating the ADCC, promising a significantly better therapeutic outcome for all patients eligible for treatment with Trastuzumab. Simultaneously, the fully human nature of Tras-GEX’ host cell lines avoids the common immunogenic reactions against host-derived non-human components.

TrasGEX has entered clinical phase I in March 2011.

Glycotope was founded in February 2001 by Dr. Steffen Goletz, CSO of Nemod Biotherapeutics and former head of a research group at the Max-Delbrueck-Centre for Molecular Medicine in Berlin, and Dr. Andreas Eckert, CEO and founder of Eckert & Ziegler Strahlen- und Medizintechnik AG.

Starting with eight employees to develop the GlycoExpress™ technology, today the company employs more than 140 people at two sites in Berlin and Heidelberg and by applying the GlycoExpress™ technology significantly widend its focus from a pure technology provider to the development of proprietary and strongly improved 2nd generation biotherapeutics. Part of this process was the acquisition of all assets of the Nemod Biotherapeutics Co. KG in 2007.

GLYCOTOPE BIOTECHNOLOGY, located in Heidelberg, comprises the former biotechnological activities of Orpegen Pharma, which Glycotope acquired in 2008. Glycotope Biotechnology allows Glycotope to produce its glycooptimized biotherapeutics under GMP conditions.

Based on Glycotope’s technology portfolio and business concepts, the participation of Hexal founders Thomas & Andreas Struengmann in 2007 constituted one of the largest biotech investments in Germany.

Glycotope is located in Berlin at the place of its foundation on the Biotech-Campus Buch in the Northeast of Berlin.

Berlin, Germany – November 12, 2010

Glycotope GmbH, a leading glycobiology company, will present recent advances with its GlycoExpressTM technology of novel human cell lines during the upcoming BIO-Europe in Munich. With two of its glycooptimized antibodies produced by GlycoExpress™ and already undergoing clinical trials, four European regulatory authorities approved GlycoExpress™ for production of biotherapeutics. This strongly validates this innovative technology for future development and production of therapeutic proteins. The preclinically observed advantages of glycooptimized biotherapeutics are expected to result in a clear clinical superiority. All glycooptimized products are produced in Glycotope´s own GMP facility in Heidelberg (Glycotope Biotechnology GmbH).

About GlycoExpress™

GlycoExpress™ is based on a tool box of glycoengineered human cell lines for glycooptimization and high yield production of glycooptimized biotherapeutics, such as antibodies. The antibody’s fully human glycosylation is optimized to yield a largely improved anti-tumor ADCC activity, bioavailability and contains no non-human immunogenic carbohydrate structures and facilitates treatment of a highly increased number of patients. Glycooptimization with GlycoExpress™ is not limited to antibodies but suitable for the majority of therapeutically relevant proteins. The technology platform is available for cooperation partners via service & license agreements.

Berlin, Germany – August 4, 2010

Glycotope GmbH, a leading glycobiology company, has received regulatory approval by German and Italian regulatory authorities for a Phase I study of Glycotope´s next generation antibody CetuGEX™ (GT-MAB 5.2-GEX) for the treatment of various solid cancers. “For Glycotope, the approval of CetuGEX™, our second antibody in the clinic, represents another important milestone,” says Steffen Goletz, CEO & CSO of Glycotope. “CetuGEX™ is our first in a series of next generation biotherapeutic products. We expect that the strong advantages in various product aspects we have seen in preclinical studies will manifest in a clear clinical superiority compared to the currently marketed product. In addition, the second cell line of Glycotope´s glycooptimization platform GlycoExpress™ based on human cell lines has now been approved, meeting our ambitions for quality and speed.” CetuGEX™ was the second antibody in clinical stage produced in Glycotope´s own GMP facility in Heidelberg.

About CetuGEX™

CetuGEX™ is an improved version of a currently marketed anti-EGFR antibody which has been approved for the treatment of colorectal and head & neck cancers.

Berlin – November 23rd, 2009

Glycotope GmbH, a leading German Biotech company, has received regulatory approval by Germany and Italian regulatory authorities for a Phase I study of Glycotope´s lead antibody GT-MAB 2.5-GEXTM for the treatment of various solid cancers. The approvals further underline the suitability of Glycotope´s proprietary GlycoExpressTM technology for the improvement, glycooptimization and high yield production of therapeutic proteins for the use in humans.

“The approval of GT-MAB 2.5-GEX as well as the regulatory approval of the GlycoExpress production technology based on its glycoengineered human cell lines represents a significant milestone for the company” says Steffen Goletz, PhD, Founder, CEO and CSO of Glycotope. “After generating very encouraging data in pre-clinical studies, we are now looking forward to demonstrate the importance of glycosylation in the clinic by generating a substantial benefit to patients.”

The Phase I study will evaluate the safety and tolerability of GT-MAB 2.5-GEXTM in a broad series of cancer indications.

About GT-MAB 2.5-GEX

GT-MAB 2.5-GEXTM is a novel, exceptionally potent monoclonal antibody for the treatment of a broad variety of cancer indications. The antibody is directed against a novel tumor-specific combined carbohydrate-protein epitope present in a large number of patients of various cancers. GT-MAB 2.5-GEXTM shows three highly effective key modes of anti-tumor action: ADCC, phagocytosis and induction of apoptosis with an exceptionally high and specific tumor accumulation and tumor killing already at doses as low as 0.5 mg/kg.

The antibody’s fully human glycosylation is optimized to yield a largely improved ADCC activity, bioavailability and no non-human immunogenic carbohydrate structures. This was achieved by Glycotope´s proprietary technology platform GlycoExpressTM, a screening and high yield production system of glycoengineered human cell-lines that allows significant enhancement of therapeutic potency by optimizing a protein’s glycosylation in various aspects.

About Glycotope GmbH

GLYCOTOPE was founded in 2001 and focuses on the improvement and humanization of glycosylation structures on proteins, a comparatively new field in biotechnology. GlycoExpressTM, the company’s proprietary technology permits making existing and new drugs more effective and tolerable in the human body, which is of considerable medical as well as economic importance. The current product range currently includes both proprietary antibodies for cancer therapy (e.g. GT-MAB 2.5-GEXTM) and significantly improved versions (2nd generation) of antibodies and other therapeutic proteins already on the market.

Heidelberg, 24 July 2009 – Progressive CMV infections pose a major threat to transplant recipients. CD8 T cells are considered as the major player to control CMV viremia and sufficient levels of CMV-specific CD8 T cells are found to protect against CMV disease.

Current assays based on PCR and antigen-detection are capable of analysing the viral burden and detect a CMV reactivation, however, any immunoreactivtity against CMV cannot be assessed.

Glyctope Biotechnology has developed a ready-to-use CMV-Tetramer Test that provides for the first time a possibility to quantify the number of CMV-specific CD8 T cells, thus helping the clinician to assess patients with high risk to develop CMV disease.

Main assay features:

Standardized and CE-certified ready-to-use staining kit (performance time about 2h)

Direct analysis of whole blood

Highly specific and sensitive detection of CMV-specific CD8 T cells due to the use of tetramers

Combination of well characterized CMV peptides that cover about 80% of HLA types

Determination of the absolute numbers of CMV-specific CD8 T cells and the percentage CD4 and CD8 T cells.

Economic Benefits:

Kit contains all necessary compounds including a negative control

Easy and economic procedure based on flow cytometry achieving quick results at low costs with high accuracy and precision

Expected benefits of monitoring the CMV-specific CD8 T cell number:

The Test allows the clinician to assess the immune status of the patient to combat with the progressive viral infection.

It helps the clinician to reduce the amount of toxic and expensive antiviral drugs in the treatment

Berlin, Heidelberg, 24 July 2008.
Glycotope Biotechnology GmbH, a fully consolidated subsidiary of Berlin-
based Glycotope GmbH, has acquired the biopharmaceutical production
facility and diagnostic business of Orpegen Pharma GmbH, which has been
based in Heidelberg for over two decades.

“With the acquisition of the production capacities in four separate GMP suites, we are creating excellent basic conditions for the further development of our glycosylation technology, with which we are improving both our own and other products already on the market,” explains Steffen Goletz, founder and scientific head of the company. In addition, GLYCOTOPE now operates independently in the GMP-based production of its development candidates. The company already has its first manufacturing licenses under the Medicines Act (AMG).

“We see this as an important step in optimising the value creation chain for the
strategic positioning of our associate company,” says Helmut Jeggle, representative of Athos GmbH. The purchase was financed and co-initiated through another capital increase signed by investor Jossa Arznei GmbH. Behind Jossa stands Athos GmbH which, as the family office of the Strüngmann brothers, now makes its know-how available to select biotech companies and embarks

“In order to further expand the capital-intensive, pioneeringbiotechnology position at the Heidelberg production site with the glycoprotein know-how developed at ORPEGEN, the transfer of biopharmaceutical assets to the financially stronger GLYCOTOPE was the logical, appropriate thing to do,” says Christian Birr, managing partner of ORPEGEN Pharma, which he founded in 1982. The development and production of peptide agents, amino acid derivatives and small molecules will be independently continued by the former Chemistry division at the site, while co- shareholder Hightech Beteiligungen GmbH & Co. KG will withdraw from its involvement in ORPEGEN Pharma.

About Glycotope GmbH
The product range of GLYCOTOPE GmbH covers various antibodies and other biopharmaceuticals. This includes both its own antibodies for tumour therapy (PankoMab, KaroMab) and much improved versions (2nd generation) of antibodies already available on the market or other proteins used in therapy.

The innovative character of GLYCOTOPE’s basic technology “GlycoExpress™”, which is based on glyco-optimised human high-production cell lines, lies in the development of new drug compounds as well as in the humanisation and optimisation of glycosylation structures and hence the associated enormousincrease in the effectiveness of so-called “2nd generation” agents.
