Antibody Technologies

Antibodies are glycoproteins which are increasingly applied in the clinic for the treatment of a broad range of diseases. Depending on the purpose antibodies are used in different formats. GLYCOTOPE offers a nearly complete range of antibody engineering technologies which we used in a series of different projects to successfully generate chimaeric and humanized antibodies (various human IgG and IgM isotypes) as well as antibody fragments including stable multibodies.
Our special humanization strategy allows us to generate a set of different functional humanized antibodies in a short time. For expression of the antibodies we use either the known rodent expression cell lines including CHO or our own human expression system GlycoExpress.

Special emphasis is taken for the preclinical evaluation and optimisation of antibodies according to their activity and specificity using a complete set of in vitro and in vivo tests. GLYCOTOPE is specialized in characterizing antibodies in cellular in vitro bioassays, like ADCC, CDC, phagocytosis, apoptosis induction, internalisation, affinity towards antigens naturally accommodated on cells, which are complemented by a series of biochemical assays, like flow cytometry, immunocytology, immunohistochemistry, ELISA, RIA, Western analysis, BiaCore. In addition, GLYCOTOPE conducts together with our local partner EPO-GmbH animal studies (rodents) including pharmacokinetc studies, biodistribution, and efficacy models.

Our large experience includes the successful development of naked antibodies, toxin-labelled antibodies and radiolabelled antibodies.
