Rituximab (2nd generation)
Rituximab is a chimeric antibody targeting CD20 and is used for treatment of patients with non-Hodgkin’s
lymphoma and Rheumatoid arthritis.
Major new indications are currently tested in Phase III.
Rituximab operates by induction of antibody-dependent cellular cytotoxicity
(ADCC) and the complement-dependent cytotoxicity (CDC) directing cytotoxic killer T cells and complement factors to kill
and destroy CD20 positive tumor cells.
Both, ADCC activity and CDC activity of an antibody depends very much on the glycosylation of an antibody at the constant region.
Antibodies expressed and GlycoOptimized in GlycoExpress show a large increase in cytotoxic activities and improvement in their
bioavailability via optimisation of their glycosylation, as demonstrated with other antibodies such as Glycotope’s
own antibody PankoMab. Additionally, GlycoExpress is a rapid high yield serum-free production system superior to current production
systems on the market.
We are currently developing a largely improved Rituximab using GlycoExpress.
